PT-141 (Bremelanotide) is a centrally acting melanocortin receptor agonist used to help support sexual desire and arousal. It is FDA-approved for hypoactive sexual desire disorder (HSDD) in premenopausal women and is also used off-label in men with low libido or erectile dysfunction, particularly where central causes are suspected. Unlike traditional PDE5 medications, PT-141 works through the central nervous system rather than by acting on blood flow.
PT-141 activates melanocortin receptors (MC3R and MC4R) in the hypothalamus, supporting sexual desire and drive through central nervous system pathways. It does not depend on nitric oxide pathways or vascular mechanisms.

Common side effects may include nausea (often transient), flushing or warmth, headache and injection-site reactions. Less common effects that should be monitored include temporary elevation in blood pressure, fatigue or yawning, dizziness, and rarely darkening of gums or skin with prolonged use. Not recommended for adults with uncontrolled hypertension or significant cardiovascular disease. Do not exceed the dosing frequency prescribed by your provider.
